    'Masha's room' in russian is komnata Mashi.
    How about 'in Marsh's room' ? V komnate Mashi ???

    It only works for the nouns of first declention when the endings are "a" or "я", regardless of a gender.

    Misha's room -Мишина комната

    Yes, -ина for feminine nouns, -ин for masculine nouns, - инo for neuter nouns. (ванина ручка, кошкин дом, катино платье.)

    Пока не прибежал alexei и не начал опять обвинять меня в том, что и эти примеры "doesn't look natural at all" скажу, что лично я перефразировал бы эти предложения так:

    Комната Игоря.
    В комнате Игоря.

    На мой взгляд, эти варианты звучат лучше. Хотя вот 'Машина комната' и 'комната Маши' - примерно одинаково.

    Grammaticaly speaking, it is allowed to say the car's room. Kwatts wanted to know how to tell the difference between the car's room and Masha's room.

    1 Машина комната
    2 комната машины

    1 can never mean the car's room, because Машина would have to be in genetive, leading to case number 2.
