The first one sounds clumsy and is hardly ever heard. The second one is perfectly fine.

"Моя семья – это 6 человек."

It means "There are 6 people in my family." right?
Literally, it means "My family is 6 persons."
If you want to say "There are 6 people in my family.", say "В моей семье 6 человек".
